The festival will take place in Hackney’s Victoria Park on Sunday 26 May, the day after Field Day.
As One will be using Field Day’s infrastructure and production company, Loudsound.
Singer Rita Ora has been announced as the headline artist. Other confirmed acts include Holly Johnson of Frankie Goes to Hollywood fame, Katy B and Union J.
There will be four large indoor arenas and five additional stages, which will be hosted by some of the world’s leading gay party brands.
Craig Elder, head of Orange Nation, said: "We felt it was time that London was no longer overlooked when it came to a major gay outdoor entertainment event. Over a year in the planning we [will] transform one of our most successful club brands into a massive outdoor event on one of London’s most loved festival sites, Victoria Park."
The As One club night has been held at Vauxhall’s Fire since 2010.
