Timberlake, who is launching his first album in six years, will make his only UK festival appearance this year when he headlines the festival on 12 July. Wireless will also host Jay Z’s only European performance of 2013 on 13 July.
The pop music festival is relocating to the Olympic Park from Hyde Park, after organiser Live Nation lost its contract for the park to AEG Live.
AEG recently announced its plans for a new series of concerts and events in the park this summer, called British Summer Time.
Steve Homer, the festival’s promoter at Live Nation, said: "Securing these two artists – Jay Z for his only European performance of 2013 and Justin Timberlake for his only UK festival appearance this year – cements our reputation as a world class player on the festival circuit."
The festival is in its ninth year and other stars revealed for the line-up include Snoop Dogg, John Legend, Frank Ocean, Emeli Sande, Rita Ora and Calvin Harris. Tickets go on sale 22 February at 9am.
